Betulinic Acid Ameliorates Cerebral Injury in Middle Cerebral Artery Occlusion Rats through Regulating Autophagy

چکیده
Cerebral ischemic stroke (CIS) is an acute cerebrovascular disease that caused by the sudden rupture of blood vessels inside brain and intervention reperfusion to brain, resulting in severe cerebral injury. Autophagy has been reported be involved occurrence progression CIS. Betulinic acid (BA) a pentacyclic triterpene mainly extracted from birch bark. Studies have shown neuroprotective effects BA. Here, effect mechanism BA on ischemia-reperfusion induced injury was explored using CIS model vivo via 1 h middle artery occlusion (MCAO) 24 rats vitro oxygen–glucose deprivation/reperfusion (OGD/R) PC12 cells, respectively. We found not only reduced reducing oxidative stress but also activated SIRT1/FoxO1 pathway suppress autophagy improve MCAO rats. These results provide basis for potential clinical application

Thrombotic Middle Cerebral Artery Occlusion in Rats
Background and Purpose: We examined the effects of the noncompetitive N-methyl-D-aspartate receptor antagonist MK-801 using a newly developed stroke model of thrombotic distal middle cerebral artery occlusion under conditions of carefully controlled head temperature. Methods: Male Sprague-Dawley rats were treated with 1 mg/kg of MK-801 or saline before the induction of ischemia. An argon laser-...
متن کاملResearch Paper: Optimization of Transient Focal Cerebral Ischemia Model by Middle Cerebral Artery Occlusion
Introduction: Cerebral ischemia is one of the most common causes of death in human populations in the industrial communities. The need for animal models is inevitable to study the pathophysiology and treatment of cerebral ischemia in human. The current study aimed at evaluating the strengths and weaknesses of different techniques used to create ischemia in previous studies and optimizing the tr...
متن کاملAttenuating of NF-Κb/VCAM-1 Expression in Middle Cerebral Artery Occlusion (MCAO) Model by Viola Odorata: Protection Against Injury Ischemia- Reperfusion Injury in Rats
Background: The death of neurons and cerebral edema are the main consequences of stroke. However, inflammatory processes play a key role in aggravating cerebral damage following stroke. The aim of this study was to investigate the effects of Viola odorant extract (VOE) on infarct volume (IV), neurologic deficits (ND), and expression of NF-κB and VCAM-1 in the MCAO model. Method: The animals we...
متن کاملReversible middle cerebral artery occlusion without craniectomy in rats.
To develop a simple, relatively noninvasive small-animal model of reversible regional cerebral ischemia, we tested various methods of inducing infarction in the territory of the right middle cerebral artery (MCA) by extracranial vascular occlusion in rats. In preliminary studies, 60 rats were anesthetized with ketamine and different combinations of vessels were occluded; blood pressure and arte...
متن کاملCorticofugal axonal degeneration in rats after middle cerebral artery occlusion.
We used the Fink-Heimer method to study degenerating corticofugal axons after unilateral middle cerebral artery occlusion in 14 adult male Long-Evans hooded rats. Axonal degeneration in the pyramidal tracts was prominent at 1-3 weeks, manifesting in well-defined silver-impregnated axonal bundles coursing from the internal capsule to the pyramids and crossing completely to the contralateral spin...
